If this is for a comic strip, you should think about the intended audience. For example, if the intended audience is Japanese, their reading direction is going to default downwards whereas an English audience's reading direction is horizontal. Because of the way you learn to read based on your language and the one that matches your natural reading direction based on that tends to be more aesthetically and sequentially pleasing since you're more likely to find it easier to read.
Book comics get a bit more complicated since you have to take into consideration how the panels interact and how your viewers' eyes will flow from panel to panel.
Comic layouts should always take the reader into consideration first, as the goal of layouts is to make it easy to read while keeping the page interesting. The second thing to take into account is the kind of story you're trying to tell. Not every comic will work well in a strip and not every comic will work in a comic book format.
